Abstract:
Bu çalışmada, poly(3-acetylthiophene) ile modifiye edilmiş camsı karbon elektrot kullanılarak ilaç örneklerinde dopamine ve norepinephrine'nin hızlı, güvenilir ve duyarlı bir şekilde saptanması için elektroanalitik yöntemler geliştirilmiştir. Poly(3-acetylthiophene) modifiye camsı karbon elektrot, 3- acetylthiophene'nin 0.05 M NaClO4 ile desteklenmiş asetonitril çözeltisinde elektropolimerizasyonu ile dönüşümlü voltammetri kullanılarak hazırlandı. pH' sı 4.0 olan 0.1 M fosfat tampon çözeltisinde (PBS), poly(3-acetylthiophene) ile modifiye edilen camsı karbon elektrot, hem dopamin ve hem de norepinefrin'nin yükseltgenmesine mükemmel bir elektrokatalitik aktivite göstermiştir. Ayrıca, modifiye elektrot dopamin ve norepinefrin'nin yükseltgenme potansiyellerinde katodik yönde önemli bir kayma ve anodik akım cevaplarında büyük bir artış gösterdi. Elde edilen sonuçlar, poly(3- acetylthiophene) ile modifiye edilmiş elektrotun aynı zamanda askorbik asit varlığında bile dopamin veya norepinefrin'nin saptanmasına uygulanabileceğini göstermiştir. Modifiye elektrot sistemi başarılı bir şekilde ilaç örneklerine uygulanmıştır.
In this study, electro analytical methods were developed for rapid, reliable and sensitive determination of dopamine and norepinephrine in pharmaceuticals using a poly(3-acetylthiophene) modified glassy carbon electrode (GCE). The poly(3- acetylthiophene) modified glassy carbon electrode was prepared by the electro polymerization of 3- acetylthiophene in acetonitrile solution containing 0.05 M KClO4 using cyclic voltammetry. The poly(3- acetylthiophene) modified glassy carbon electrode exhibited an excellent electro catalytic activity towards the oxidation of both dopamine and norepinephrine. The modified electrode showed a remarkable shift of the oxidation potentials of both dopamine and norepinephrine in the cathodic direction and a drastic enhancement of anodic current response in 0.1 M phosphate buffer solution (PBS) at pH 4.0. The results obtained indicated that the poly(3-acetylthiophene) modified electrode could also be applied for the determination of dopamine or norepinephrine in the presence of ascorbic acid. The modified electrode system has successfully been applied to the pharmaceutical formulations.
