Abstract:
Bu araştırma, 2007-2008 yılları arasında Harran Üniversitesi Ziraat Fakültesi Laboratuvarlarında yürütülmüştür. Çalışmada materyal olarak domates bitkisi, solgunluk fungusu Verticillium dahliae ve farklı konsantrasyonlarda NaCl kullanılmıştır. Denemeler saksılarda 3 tekerrülü olarak tesadüf parselleri deneme desenine göre kurulmuştur. Domatesler yetiştirildikten sonra bir gruba kök-daldırma yöntemi ile V. dahliae inokule edilmiş, diğer gruba ise fungus ile birlikte 50, 100, 150, 200 ve 250 mM NaCl uygulanmış, kontrol grubu ise sadece fungus ile inokule edilen veya ne fungus ne de tuz ile muamele gören grublardan oluşmuştur. Bitki gelişiminin yanı sıra, biyokimyasal metabolitlerden peroksidaz, polifenoloksidaz, katalaz ve prolin ölçülmüştür. Fungus ile inokule edilen ve artan tuz konsantrasyonlarına maruz bırakılan bitkiler, kontrol grubu bitkilere kıyasla gelişme geriliği göstermiş, buna bağlı olarak biyokimyasal metabolitler de etkilenmiştir
This research was carried out in 2007-2008 in the laboratories of Faculty of Agriculture in Harran University. In the study, tomato plants, the wilt fungus Verticillium dahliae and salt were used as materials. The trial was set up in a randomized block design with three replicates. After the growth of tomato, one group was inoculated with root-dipping method with V. dahliae and the other group was inoculated with V. dahliae followed by the treatments of varios concentrations of NaCl (50, 100, 150 and 200 mM). As control groups only inoculated plants with V. dahliae or the plants with no treatment were used. Apart from plant development criteria, some biochemical parameters such as peroxidase polyphenoloxidase, catalase, and proline were measured. Inoculated plants with the fungus followed by exposed to varios concentrations of NaCl showed drastic decline in growth parameters and their biochemical parameters were aso affected compared to the control groups.
