Muz (Musa sp.) klonlarının doku kültüründe farklı maddeler kullanılarak üretilmesi / Propagation of bananaclones (Musa sp.) using different gelling agents under in vitro conditions

Abstract

Bu çalışma Harran Üniversitesi Biyoloji Bölümü ve Gazi Üniversitesinin Biyoloji Eğitim Anabilim Dalı'nın Biyoteknoloji Laboratuvarında ortak olarak yürütülmüştür. Türkiye de ekonomik önemi yüksek olan bitki türlerine yönelik doku kültürü çalışmaları son yıllarda hız kazanmıştır. Klasik muz çoğaltımda kullanılan üretim materyalleri homojen olmamakta ve bunların hastalık ve zararlılarla bulaşık olma riski de oldukça yüksek olmaktadır. Bu sebeplerden dolayı muz çoğaltımında doku kültürü ile çoğaltılmış fideler yavaş yavaş çiftçiler tarafından büyük ilgi görmektedir. Ancak maliyeti yüksek olan bu fideler tüm çiftçiler tarafından kullanılamamaktadır. Bu sorunun muz doku kültürü teknolojisindeki ucuz katılaştırıcı maddeler ve karbon kaynakları kullanarak çözülebileceği düşünülmüştür. Denemelerde kullanılan Dwarf Cavendish muz çeşidi Alanya'dan temin edilmiştir. Eksplant olarak muz meristemi kullanılmıştır. Çalışmada karbon kaynağı olarak sukroz ve ticari beyaz şeker ve katılaştırıcı madde olarak ta agar, isubgol ve pamuk lifleri kullanılarak muz doku kültürü maliyeti düşürülmeye çalışılmıştır. Agar'a göre isubgol, pamuk lifleri ve Sukroz'a (Sigma) göre beyaz şeker oldukça ucuzdur. Çalışmada muz rizom eksplantları, steril edilip 50 mg/l askorbik asit içeren sıvıda yarım saat bekletildikten sonra, değişik katılaştırıcı madde ve sukroz ile ticari beyaz şeker içeren 4.5 mg/l BAP ve 0.2 mg/L IBA bulunduran MS ortamında kültüre alınmıştır. Tüm katlaştırıcı madde, sukroz, ticari beyaz şeker 4.5 mg/l BAP ve 0.2 mg/L IBA içeren MS ortamları arasında istatistiksel olarak farklılık bulunmamıştır. Tüm katılaştırıcı maddeler üzerinde gelişen sürgünler, 0.2 mg/l IBA içeren MS ortamlarında köklendirilip adaptasyon sağlanmıştır. Çalışma sonucunda sabit masraflar hariç, agar- sukroz içeren ortamda en pahalı, pamuk ticari- beyaz şeker içeren ortamda ise en ucuz bitki elde edilmiştir. Elde edilen bulgular teknolojinin gelişmesine yardımcı olacak ve aynı zamanda yurt dışına giden döviz miktarını da azaltacakt The study was jointly conducted at the Harran University and the biotechnology laboratory of the department of Biyology education of Gazi University, Ankara. Tissue culture of economically important plants has gained considercable importance during last few years. The traditional propagation methods are not homogenous and have high risks of carrying diseases and pests. Due to this reason the plantlets produced through tissue culture are gradually becoming popular among farmers. However, due to high cost all farmers have not approach to these plantlets. To reduce the cost, the study planned to solve this problem by using different solidifying matters and carbons sources. The plant material was obtained from private farms of Alanya and banana meristem was used as explant. Agar, isubgol, and cotton fibres were used as solidifying agent and commercial white sugar and sucrose were used as carbon source in this study. Compared to agar, isubgol and cotton fibres and compared to Sigma sucrose commercial sugur are very cheap. After sterilsation, the banana rhizome exsplants were treated with 50 mg/l ascorbic acid for ½ hour and thereafter cultured on MS medium supplemented with 4.5 mg/l BAP-0.2 mg/l IBA. No statistical difference was observed in number of shoots obtained on all solidifying media and carbon sources. These were rooted on respective MS containing 0.2mg/l IBA and adapted. Excluding fixed expenditures, medium containing agar-sugar was the most expensive and the medium containing cottonwhite commercial sugar was the cheapest to propagate banana plantlets. These results will help for the deveolpment of technology and at the same time will help in saving the foreign exchange that goes abroad.
