Abstract:
Bu çalışmada Beyaz peynir üretiminde alternatif bir pıhtılaştırıcı olarak rekombinant kimozin kullanım olanakları araştırılmıştır. Bu amaçla A: ticari buzağı renneti, B: rekombinant kimozin (Aspergillus n iger var. awamori) ve C: mikrobiyel rennet (Rhizomucor m ieheî) kullanılarak Beyaz peynir üretilmiş ve örnekler %12'lik salamurada 4±1 °C'de 60 gün süreyle depolanmıştır. İki tekerrürlü olarak yürütülen bu çalışmada depolamanın 1., 15., 30. ve 60. günlerinde pH, titrasyon asitliği, kurumadde, yağ, kurumaddede yağ, protein, tuz, kurumaddede tuz, azotlu bileşenler (toplam azot, suda çözünen azot, olgunlaşma indeksi, protein olmayan azot, proteoz-pepton azotu ve kazein azotu) elektroforetik ve duyusal özellikler belirlenmiştir, örneklerin duyusal değerlendirmeleri 10 kişilik bir panel tarafından yapılmıştır. Analizlerden elde edilen sonuçlara göre, peynirlerin titrasyon asitliği, kurumadde, tuz ve kurumaddede tuz, suda çözünen azot, olgunlaşma indeksi ve protein olmayan azotlu bileşikler ile koku hariç tüm duyusal özellikleri üzerine enzim çeşidinin etkisi önemli olmuştur (p<0.05). Depolama süresi boyunca peynirlerin incelenen tüm özelliklerinde önemli farklılıklar bulunmuştur (p<0.05). Depolamanın sonunda başlangıç değerlerine göre titrasyon asitliği, tuz, kurumaddede tuz, suda çözünen azot, olgunlaşma indeksi ve protein olmayan azot içerikleri ile duyusal puanlan artarken, pH, yağ, toplam azot, protein ve ccsl-kazein içerikleri ise azalmıştır. In this study, the possibilities of using recombinant chmosin as an alternative coagulant in White cheese production was investigated. For this purpose, White cheese produced by using A: commercial calf rennet, B: recombinant chymosin (Aspergillus niger var. awamori) and C: microbial rennet (Rhizomucor miehei). The cheese samples were stored in % 12 brine at 4±1 °C for 60 days. In the study, which was carried out in duplicate, pH, titratable acidity, dry matter, fat, fat in dry matter, protein, salt, salt in dry matter, nitrogeneous compounds (total nitrogen, water soluble nitrogen, ripening index, non-protein nitrogen, proteos-pepton nitrogen and casein nitrogen), elektrophoretic and organoleptic properties of cheese samples were determined in 1., 15., 30. and 60. days of storage. Organoleptic evaluations were achieved by a panel consisting 10 panelists. According to the results, the effects of enzyme type was significant on the titratable acidity, dry matter, salt, salt in dry matter, water soluble nitrogen, ripening index, proteos-pepton nitrogen and all sensory properties, except for the smell (p<0.05). Significant differences were observed in all properties of the cheese samples except for fat-in-dry matter (p<0.05). At the end of storage, the titratable acidity, salt, salt in dry matter, water soluble nitrogen, ripening index, non-protein nitrogen values and sensory scores of the cheeses increased, while the pH, fat, total nitrogen, protein and ctsi-casein contents of cheeses were decreased compared to initial values.
