Abstract:
Fungal etmenler patojenisitesini penetrasyon kabiliyetine, salgıladıkları hücre dışı enzimlere ve toksinlere borçludurlar. Özellikle proteinlerin yapısını bozan proteaz enziminin fungal etmenler tarafından salgılanmaları onların karakterleri hakkında bilgi verir. Penicillium expansum fungusunun biyokimyasal olarak patojenik karakterini ortaya çıkarmak için farklı sıcaklıklardaki sporulasyonu ve proteaz ve lakkaz enzimlerinin durumu incelenmiştir. Fungal etmen, hem düşük (4-8 °C) hem de yüksek sıcaklık olarak kabul edilen 35 °C ve üstü sıcaklıklarda biyokimyasal faaliyetlerine devam etmiş, 55 °C'de ise enzimsel ve diğer fizyolojik faaliyetleri son bulmuştur. Bu etmenin hasat sonrası meyve ve sebzelere kontaminasyonu durumunda sıcaklık ayarlaması ile kontrol edilemeyeceği, ilave tedbirlere gereksinim duyulduğu anlaşılmıştır. Pestisit kullanımının sınırlandırıldığı bir dönemde alternatif olarak öne sürülen yöntemlerin etkinliğinin belirlenmesinde bu çalışmada kullanılan parametrelerin önemli kriterler arasında sayılabileceği belirlenmiştir.
Pathogenicity of fungal agents depends on the penetration ability, production of extracellular enzymes and toxins. Secretion of protease enzymes, degrading of protein structures, by fungal agents would inform us about the pathogenic characteristics of the pathogens. Production levels of protease and laccase enzymes and sporulations of Penicillium expansum fungus at different temperatures were elucidated. The fungal agent carried out its physiological and biochemical functions both at low (4-8 °C) and high (35 °C and above) temperatures, enzymatic and other physiological functions were ended at 55 °C. It was established that this fungal agent could not be controlled via adjustment of temperature at storage conditions in case of contamination of fruits and vegetables, in fact, further control measures are needed to control the pathogen. In the era of pesticide restriction, these parameters used in this study could be counted as important criteria and proposed to test the efficacy of methods alternatively suggested.
