Abstract:
Trichophyton verrucosum, sığır dermatofitozuna neden olan en önemli etkendir. Deri lezyonları, genç sığırlarda gelişmede gerileme, süt sığırlarında sütte azalma, derinin ekonomik değerinin azalması gibi önemli sorunlara neden olmaktadır. Etken tanımlamada direkt mikroskobik inceleme ve kültür, geleneksel konvansiyonel yöntemlerdir. Standart konvansiyonel yöntemler zaman alıcı olması ve kesin sonuçlar vermemesinden dolayı duyarlılığı ve özgüllüğü yüksek, hızlı yeni tanı yöntemlerine ihtiyaç bulunmaktadır. Bu çalışmada sığırlardan Trichophyton verrucosum izolasyonu ve MALDI TOF-MS ile doğrulanması amaçlanmıştır. Dermatofitoz şüpheli 100 adet sığırdan alınan deri kazıntısı ve kıl örnekleri direkt mikroskopi yöntemi ile incelenmiş ve 52 (%52)'si pozitif bulunmuştur. Numunelerin kültür yöntemi ile incelenmesi sonucunda 63(%63)'ünde dermatofit izole edilmiş ve bunların 52 (%52)'si T.verrucosum olarak identifiye edilmiştir. Bütün Trichophyton verrucosum izolatlarının 7950-7954 arasında MALDI TOF-MS spektrum pikleri saptanarak, doğrulanmıştır. Sonuç olarak, Şanlıurfa bölgesinde sığırlarda T. verrucosum'un yaygın olduğu, MALDI TOF-MS tekniğinin T.verrucosum'un teşhisinde geleneksel yöntemlere göre başarılı bir şekilde kullanılabileceği belirlendi. Trichophyton verrucosum, is the most important causative agent for cattle dermatophytosis. It causes skin lesions, growth retardation in young cattle, decline in milk production, reduction economic value of leather. Direct microscopy and culture are conventional methods for agent identification. Standard conventional methods are time consuming and they might not be very definitive and therefore it is needed the new and fast diagnostic techniques that have high sensitivity and specificity. In this study, the isolation of Trichophyton verrucosum from cattle and confirmation with MALDI TOFMS of the isolated strains were aimed. Skin scrapings and hair samples collected from 100 were examined by direct microscopy and 52 (52%) of them were found as positive. At the end of cultural studies, 63 (63%) dermatophytosis agents were isolated and 52 of them were identified as T.verrucosum. All Trichophyton verrucosum isolates showed MALDI TOF-MS peaks between 7950 and 7954 and these results confirmed the Trichophyton verrucosum isolation. As conclusion, it was determined that Trichophyton verrucosum infection was common in Şanlıurfa Region and MALDI TOF-MS technique could be used sucessfully compared to conventional techniques for diagnosis of T.verrucosum.
