Abstract:
Trigonella cinsi, yaygın olarak yetiştirilen Trigonella foenum-graecum ve Akdeniz bölgesi çevresinde dağılmış diğer bazı tıbbi ve aromatik bitki türlerini içerir. Trigonella türlerinin sınıflandırılması morfolojik bulgulara dayandırıldığından dolayı türler arası sınıflandırmalar henüz tam olarak çözülememiştir ve taksonomik sorunlar bulunmaktadır. Moleküler tekniklerin gelişmesi ie birlikte taksonomik çalışmalarda da çeşitli teknikler kullanılmaktadır. Bu çalışma ile Trigonella cinsinde yer alan Bucerates ve Foenum-graceum seksiyonlarına ait 49 örneğin, moleküler bir teknik olan Rastgele Çoğaltılmış Polimorfik DNA-Polimeraz Zincir Reaksiyonu (RAPD-PCR) yöntemi ile tür içi ve türler arası akrabalık ilişkilerinin ortaya konulması amaçlanmıştır. Analiz için 20 adet RAPD primeri denenmiş ve bunlardan 13 tanesinden tekrarlabilir amplifikasyonlar elde edilmiştir. Yapılan çalışmalar sonucu 13 primerden 222 farklı bant elde edilmiş olup, bu bantların tamamı (%100) polimorfiktir. En fazla bant sayısı OPP09 (23), en az bant sayısı ise OPB08 (13) primerleinden elde edilmiştir. Kümeleme analizi için UPGMA (Unweighted Pair Group Method using the Arithmetic Average) algoritması kullanılarak elde edilen dendrograma göre, çalışılan 49 örnek 6 ana gruba ayrılmıştır. Bu araştırma, RAPD tekniğinin farklı Trigonella türleri arasındaki ilişkinin belirlenmesi ve tür içi polimorfizmin değerlendirilmesinde yararlı bir yöntem olduğunu göstermiştir.
The genus Trigonella includes the commonly cultivated Trigonella foenum - graecum and some other medicinal and aromatic plant species distributed around the Mediterranean region. Because of the classification of Trigonella species is still based on morphological findings, in-species classification has not yet been fully resolved and there are taxonomic problems. Along with the development of molecular techniques, various techniques are used in taxonomic studies. In this study, it was aimed to reveal relations between different species and intraspecific relationships of Bucerates and Foenum-graceum sections of Trigonella genus by using Random Amplified Polymorphic DNA-Polymerase Chain Reaction (RAPD-PCR) method which is a molecular technique. For the analysis, 20 RAPD primers were tested and reproducible amplifications were obtained from 13 primers. As a result of the present study, 222 different bands were obtained from 13 primers, all of these bands were polymorphic (%100). The maximum number of bands was obtained from OPP09 and the minimum number of bands was obtained from OPB08 primers. According to the dendrogram obtained using the UPGMA (Unweighted Pair Group Method using the Arithmetic Average) algorithm for clustering analysis, 49 samples were divided into 6 main groups. This research showed that the RAPD technique is a useful tool for determining the relationship between different Trigonella species and evaluating intraspecific polymorphism.
