Diyarbakır ili ve ilçelerinde üretilen kabakgillerdeki viral etmenlerin tanılanması ve yaygınlıklarının belirlenmesi / Determination and incidence of viruses in cucurbits grown in Diyarbakır and its districts

Abstract

Bu çalışma, 2014 yılında Diyarbakır ili Merkez, Bismil, Çınar, Dicle, Eğil, Ergani, Hani ve Silvan ilçelerinde kabakgil üretiminin yoğun olarak yapıldığı 24 köyde yapılmıştır. Virüs simptomu gösteren 34 adet hıyar, 176 adet kabak, 142 adet karpuz ve 195 adet kavun olmak üzere toplam 547 adet örnek, viral etmenlerin tanılanması ve yaygınlıklarının belirlenmesi amacıyla alınmıştır. Cucumber Mosaic Vırus (CMV), Watermelon Mosaic Virus 2 (WMV–2), Squash Mosaic Vırus (SqMV), Zucchini Yellow Mosaic Vırus (ZYMV), Papaya Ringspot Virus (PRSV), Cucurbite Aphid-Borne Yellows Virus (CABYV) ve Melon Necrotic Spot Virus (MNSV)'e karşı Double-antibody sandwich enzyme linked immuno sorbent assay (DAS-ELISA) yöntemi uygulanarak testlenmiştir. Yapılan testler sonucunda 547 örnekten 100'ü CMV, 261'i WMV, 101'i SqMV, 152'si ZYMV, 81'i PRSV, 17'sinin CABYV ile bulaşık olduğu saptanmıştır. Buna göre; CMV %18.28, WMV % 47.71, SqMV %18.46, ZYMV %27.78, PRSV %14.8 ve CABYV %3.10 oranında bulunmuştur. Yapılan araştırmada hiçbir örnekte MNSV infeksiyonuna rastlanmamıştır. This study was conducted in 2014, in 24 different villages of Bismil, Cınar, Dicle, Egil, Ergani, Hani, and Centre districts of Diyarbakır, where cucurbit production is extremely high. 34 samples of cucumber, 176 zucchinis, 142 watermelons and 195 cantaloupe samples, which make 547 samples in total were collected to diagnose viral factors and to determine their frequencies. Tests were completed by applying Double- antibody sandwich enzyme linked immuno sorbent assay (DAS-ELISA) method against Cucumber Mosaic Virus (CMV), Watermelon Mosaic Virus (WMV), Squash Mosaic Virus (SqMV), Zucchini Yellow Mosaic Virus (ZYMV), Papaya Ringspot Virus (PRSV), Cucurbite Aphid- Borne Yellows Virus (CABYV) and Melon Necrotic Spot Virus (MNSV). Results from the tests applied, it was found out that 100 out of 547 samples were contaminated with CMV, 261 of it contaminated with WMV, 101 of it contaminated with SqMV, 152 of it contaminated with ZYMV, 81 of it contaminated with PRSV and 17 of the total number of samples contaminated with CABYV. Rates concluded from the results are as the following; 18.28% of CMV, WMV 47.71%, SqMV 18.46%, ZYMV 27.78%, PRSV 14.8% and CABYV 3.10%. MNSV was not found in any sample in this research.
