Abstract:
Giriş: Organ korunumuna iliĢkin modern yaklaĢımlar geliĢtirilmesine rağmen tıbbi anlamda torakoabdominal aorta cerrahisinde iskemi reperfüzyon hasarının önlenmesine yönelik geliĢmelere ihtiyaç duyulmaktadır. Bu anlamda cerrahlar bahsedilen hasarı önlemek için çeĢitli yöntemler geliĢtirse de bu yöntemlerden hiç biri tek baĢına yeterli gelmemektedir. Amaç: Bu çalıĢmada amaç timokinon, silimarin ve curcuminin, torakoabdominal aorta cerrahisiyle iliĢkili iskemi reperfüzyon hasarına karĢı antioksidant etkilerini belirlemektir. Materyal ve Metod: Bu amaçla, toplam 25 Wistar albino ırkı sıçan her bir grupta 5 sıçan olacak Ģekilde sham, kontrol ve tedavi gruplarına ayrılmıĢtır. Ġnfrarenal aortada 120 dakika boyunca iskemi uygulanmıĢ, reperfüzyon baĢlamadan peritonal açıklıktan 20 mg/kg timokinon, 200 mg/kg silimarin ya da 200 mg/kg curcumin verilmiĢtir. 60 dakikalık reperfüzyon uygulaması sonunda sağ ventrikülden alınan kan dokusunda TAS, TOS ve OSĠ ticari kitlerle fotometrik yöntemlerle, gastroknemius kas dokusunun histopatolojisi ise mikroskopik yöntemlerle araĢtırılmıĢtır. Bulgular: Timokinon etkisinde oksidatif stres indeksi azalırken TAS ve TOS değiĢmemiĢtir. Silimarin etkisinde TAS, TOS ve OSĠ azalmıĢtır. Curcumin TAS ve TOS'ta artıĢa neden olurken OSĠ'de herhangi bir değiĢiklik belirlenmemiĢtir. Gastroknemius kas dokusunda ise herhangi bir patoloji tespit edilmemiĢtir. Sonuç: Sonuç olarak, deneysel aortik iskemi reperfüzyon modeli deneyinde TAS, TOS ve OSĠ'de meydana gelen değiĢikliklere bağlı olarak kullanılan ajanların terapötik etkilerinin olduğu düĢününülmektedir. Background: Despite the developing of modern approaches for organ protection it is still needed to develop thoracoabdaminal aorta surgery operations and to protect ischemia reperfusion injury. In this sense, even surgeons developed some methods to prevent that damages, however none of these methods are sufficient by itself. Aim: Aim of this work is to determine antioxidant effect of thymoquinone, slymarin and curcumin on ischemia reperfusion injury that related with thoracoabdaminal aorta surgery. Material and Method: Totally, 25 Wistar albino rats were divided into the sham, control and treatment groups, each of those included 5 rats. Ischemia applied to the Infrarenal aorta during thoracoabdaminal aorta surgery for 120 minutes. Before the reperfusion, 20 mg/kg Thymoquinone, 200 mg/kg Slymarin or 200 mg/kg Circumin were given to the goups from peritonal orifice of the rats. After application of reperfusion for 60 minutes, the whole blood was taken from right ventricles. Total antioxidant status (TAS), total oxidant status (TOS) and oxidative stress index (OSI) in the serum derived from whole blood were studied with available commercial kits. Histopathology of the Gastrocnemius muscle tissue is investigated by microscopic methods. Findings: Oxidative stress index levels in Thymoquinone group decreases (p<0.05) while TAS and TOS levels remain unchanged (p>0.05). TAS, TOS and OSI levels decrease in Slymarin group (p<0.05). Circumin leads to increase in TAS and TOS levels (p<0.05) though there is no change detectable in OSI levels (p>0.05). There is no pathological finding in the gastrocnemius muscle tissue. Conclusion: As a result, on the experiment of aortic ischemia reperfusion model, it maybe assumed that thymoquinone, slymarin and curcumin have therapeutics effects on ischemia reperfusion injury regarding to TAS, TOS and OSI levels.
