Hedysarum aucheri (Fabaceae) bitkisinin kimyasal içeriğinin araştırılması ve farklı ekstrelerinin antioksidan aktivitesinin incelenmesi / Investigation of chemical composition of Hedysarum aucheri (Fabaceae) and evaluation of antioxidant activity of its different extracts

Abstract

Kurutulduktan ve öğütüldükten sonra Hedysarum aucheri (Fabaceae) bitkisi kimyasal olarak incelendi. Kurutulmuş materyalin sahip olduğu kimyasal içerik metanol ile bitkiden ekstrakte edildi. Elde edilen kaba metanol ekstresi daha sonra seçici katı-sıvı ve sıvı-sıvı ekstraksiyonları ile farklı ekstrelere ayrıldı. Kaba metanol ekstresi sırasıyla heksan ve diklorometan ile yıkanarak (katı-sıvı ekstraksiyonu) bu çözücülere ait ekstreler elde edildi. Geri kalan kısım üzerine bütanol ve su eklenerek (sıvı-sıvı ekstraksiyonu) maddenin tamamı bu iki fazlı karışım içerisinde çözündü ve ayırma hunisinde bu fazlar ayrılarak bütanol ve su ekstreleri elde edildi. Elde edilen bu ekstrelerin antioksidan aktiviteleri; toplam flavonoid içeriği, toplam fenolik içeriği, CUPRAC bakır(II) indirgeme esaslı antioksidan kapasite ve ABTS radikal süpürme kapasitesi testleri ile belirlendi. Ayrıca heksan ekstresinin metil esterleri hazırlanarak GC-MS analizleri yapıldı ve bitkinin sahip olduğu yağ asitleri tayin edildi. After dried and ground, Hedysarum aucheri (Fabaceae) plant was chemically examined. The chemical content of the dried material was extracted with methanol. The resulting crude methanol extract was then separated into different extracts by selective solid-liquid and liquid-liquid extractions. Extracts of hexane and dichloromethane were obtained by washing the crude methanol extract with these solvents respectively (solid-liquid extraction). By adding butanol and water over the remainder (liquid-liquid extraction), all of the material was dissolved in this two-phase mixture and these phases were separated in the separation funnel to obtain butanol and water extracts. Antioxidant activities of these extracts obtained were determined by total flavonoid content, total phenolic content, antioxidant capacity based on CUPRAC copper (II) reduction and ABTS radical sweeping capacity tests. In addition, methyl esters of hexane extract were prepared and analyzed by GC-MS, and fatty acids of the plant were determined.
