Abstract:
Bu çalışma ile İstanbul İli Anadolu yakasında bulunan 14 ilçesindeki içme ve kullanma sularında E.coli O:157 varlığı araştırılmıştır. Çalışmada 1 Nisan 2015 ile 1 Ağustos 2015 tarihleri arasında toplam 1875 adet içme ve kullanma suyu numunesi toplanmıştır. Koliform bakterilerden olan E. coli O:157'nin araştırılması için membran filtrasyon metodu kullanılmıştır. Her bir numuneden 100 ml'lik porsiyon 0,45μm gözenekli selülöz membran filtrelerden süzüldükten sonra E. coli analizi ilk olarak Tergitol besiyerinin bulunduğu petrilere yerleştirilmiştir. Petri kutuları 44 °C'de 24 saat inkübasyona bırakılmıştır. İnkübasyon sonrası sarı ve turuncu koloniler E.coli olarak kabul edilmiştir. Tergitol besiyerine ekildikten sonra şüpheli olarak görülen numuler TSA besiyerine ekilmiş ve 36±2 °C'de etüve alınıp 24 saat inkübasyona alınmıştır. İnkübasyon sonrası Oksidaz testi yapılmıştır. Oksidaz testi negatif olan numuneler, Lauryl Sulfat Broth (MUG'lu) besiyerine ekilmiş ve 44 °C'de 24 saat inkübasyona alınmıştır. İnkübasyon sonrası UV test lamba (366 nm) altında floresan göstermeyenler E. coli olarak şüpheli kabul edilmiş ve Kovak ayıracı (İndol) uygulamasıyla doğrulama yoluna gidilmiştir. Kovaks ayıracı (İndol) uygulanan numelerde pembe-mor halka oluşumu gözlenen numuneler E. coli olarak kabul edilmiştir. Yapılan analiz sonuçlarına göre toplanan 1875 adet içme ve kullanma su numunesinin 30 adedinde (%1.6) E. coli tespit edilirken, E. coli O:157 serotipi tespit edilememiştir.
