Abstract:
Kanser, ölüm nedenleri arasında ön sıralarda yer almakta ve tedavisinde de ciddi güçlüklerle karşılaşılmaktadır. Kanser tedavi yöntemlerinin en önemlilerinden biri kemoterapi olmakla birlikte henüz kesin bir tedavi metodu bulunmamaktadır. Bu nedenle yeni kemoterapotik ajan arayışları hızla devam etmektedir. Bu çalışmada, yeni kemoterapotik ajan arayışlarına katkıda bulunmak amacıyla sterik engelli fenol türevli Schiff bazlarından olan (2,4-di-tert-butil-6-{[(2,5-dimetilfenil)imino]metil}-fenol), (2,4-di-tert-butil-6-{[(3,4-metilfenil)imino]metil}-fenol), (2,4-di-tert-butil-6-{[(2,3-dimetilfenil)imino]metil}-fenol), (2,4-di-tert-butil-6-{[(3,5-dimetilfenil)mino] etil}-fenol) bileşiklerinin miyeloid kanser hücre soyu olan K562 hücre serisi üzerindeki sitotoksik etkilerinin belirlenmesi hedeflenmiştir. Bileşiklerin gösterdiği sitotoksisite düzeyleri kolorimetrik MTT yöntemiyle belirlenmiştir. Test edilen bileşiklerden gözlenen sitotoksik etki düzeyleri incelendiğinde bu bileşiklerin kemoterapotik ajan olabilmek için yeterli düzeyde etkinliğe sahip olmamakla birlikte farklı kanser türleri üzerinde test edilmesinin yararlı olabileceği fikrine varılmıştır. Cancer is a leading cause of death and serious difficulties are encountered during treatment of cancer. Chemotherapy is one of most significant treatment methods of cancer but still there is not a definite treatment method. That?s why new chemotherapeutic agent researches continue at a fast pace. In this study, with the aim of contributing new chemotherapeutic agent researches, it was aimed to determine cytotoxic effect of Schiff bases (2,4-di-tert-butyl-6- {[(2,5 ? dimethylphenyl )imino] methyl} ?phenol),(2,4-di-tert-butyl-6- { [ (3,4-methylphenyl) imino] methyl } ? phenol), (2,4-di-ter-butyl-6- {[ 2,3 dimethylphenyl) imino]methyl}-phenol), (2,4-di-tert-butyl-6-{[ [ (3,5-dimethylphenyl)mino] ethyl}-phenol) which are sterically hindered and derived from phenol, on K562 cell line of myeloid cancer cell strain. Cytotoxicity levels of compounds was determined by colorimetric MTT method. When the observed cytotoxic effect levels of tested compounds are investigated, it is decided that however these compounds don?t have sufficient level of activity to be a chemotherapeutic agent, it will be useful to test them on different types of cancer.
