Abstract:
Bu çalışmada, farklı oranlarda mikrobiyaltransglutaminaz (MTGase) enzimi ilavesinin yarım yağlı ayranların fizikokimyasal, duyusal, mikrostrüktürel ve mikrobiyolojik özellikleri üzerine etkileri araştırılmıştır. Bu amaçla ayranlara 0 ( kontrol: E), 0,5 (A), 1 (B), 2 (C) ve 4 (D) unit/g protein oranlarında MTGase enzimi ilave edilmiş ve 20 günlük depolama süresince ayranların pH, titrasyon asitliği, viskozite, serum ayrılması, su tutma kapasitesi, tat-aroma, kıvam, genel kabul edilebilirlik, Str. Thermophilus ve L. delbrueckii subsp. bulgaricus sayıları belirlenmiştir.Farklı oranda enzim ilavesinin ayranların incelenen tüm özelliklerine, depolamanın süresinin de ayranların titrasyon asitliği, viskozite, serum ayrılması, su tutma kapasitesi, Str. Thermophilus ve L. delbrueckii subsp. Bulgaricus üzerine etkileri önemli bulunmuştur (p<0,01).MTGase kullanımı ayranların fizikokimyasal, mikrobiyolojik ve kısmen de duyusal özellikleri üzerinde olumlu etki yaratmıştır. Elde edilen verilere göre duyusal olarak en iyi örnek 1 unit/gr protein oranında enzim katkılı B örneği olsa bile, diğer özellikler açısından en iyi örneğin 2 unit/gr protein oranında enzim katkılı C örneği olduğu belirlenmiştir. Sonuç olarak; yarım yağlı ayran üretiminde 1 ve/veya 2 unit/g protein oranında MTGase enzimi kullanılması önerilebilir.
In this study, the effects of microbial transglutaminase (MTGase) enzyme addition at different rates, on physicochemical, sensorial, microstructural and microbiological properties of skim ayran are investigated. For this purpose, MTGase enzyme was added to the ayran at 0 ( control: E), 0.5 (A), 1 (B), 2 (C) and 4 (D) unit/g protein rates and pH, titratable acidity, viscosity, syneresis property, water-holding capacity, taste-aroma, consistence, general acceptability, numbers of Str. Thermophilus and L. delbrueckii subsp. Bulgaricus in the ayran samples were determined for the storage period of 20 days.The effects of enzyme addition at different rates were found significant for all properties of the examined ayran; and effect of the storage time was significant for the,viscosity, syneresis, water-holding capacity numbers of Str. Thermophiles and L. delbrueckii subsp. Bulgaricus (p<0,01).The use ofMTGase enzyme created a positive effect on physicochemical, microbiologic properties and partially sensitive properties of the ayran. According to achieved data, even though the best sample is B sample which has enzyme content of 1 unit/gr protein sensorially, it was determined that the best sample is C sample which has enzyme on 2 unit/gr protein rate in terms of other properties. As a result, MTGase enzyme usage of 1 and/or 2 unit rate can be suggested for the production of reduce ayran.
