Abstract:
Bu çalışma, bazı kaba yemlerin by-pass (BP) değerlerinin in vitro enzim tekniği ile belirlenmesinde kullanılabilecek en uygun enzim konsantrasyonunun belirlenmesi amacıyla yürütülmüştür. Araştırmada yem materyali olarak 7 farklı kaba yem kaynağı kullanılmıştır. Bu yemler; fiğ kuru otu (F), buğday silajı (BSL), mercimek samanı (MS), buğday kuru otu (BKO), tritikale otu (T), yonca kuru otu (Y) ve mısır silajı (MSL)'dan oluşmuştur. Fiğ kuru otu, buğday kuru otu ve mısır silajının in sitü BP değerleri ile in vitro BP değerleri istatistiksel olarak benzer bulunmuştur (P>0.05). Buğday silajı, mercimek samanı ve yonca otunun in vitro BP değerleri in sitü değerlerinden farklı bulunmuştur (P<0.05). İn vitro teknikte inkübasyon süresinin 72 saate çıkarılması sadece fiğ ve yonca otunun BP değerlerini etkilerken (P=0.01), diğer 5 kaba yemin BP değerleri üzerine etkisi bulunmamıştır (P>0.10). Yine benzer şekilde, enzim konsantrasyonunun artırılmasının BP değerleri üzerine etkisi tüm kaba yemler için aynı olmamıştır. Bundan dolayı her yem için ayrı ayrı inkübasyon süresi ve enzim konsantrasyonun in vivo ve in sitü veriler esas alınarak, belirlemesine ihtiyaç duyulmaktadır.
This study was conducted to determine the optimum enzyme concentration of protease solution in in vitro method for determining by-pass protein value of roughages. Total 7 roughages were used as roughage sources. These roughages included vetch hay, wheat silage, wheat hay, lentil straw, triticale hay, alfalfa hay and corn silage. By pass protein values of vetch hay, wheat hay and corn silages were found similar in both in vivo and in in vitro enzyme technique (P>0.05). However, other roughages had differed BP values according to technique (P<0.05). While increasing incubation length from 48 h to 72 h affected BP values of alfalfa hay and vetch hay (P=0.01), BP values of other roughages were not affected (P>0.10). Similarly, increasing enzyme concentration did not changed BP values of all roughages. Therefore, future studies should be focused on enzyme concentration and incubation length for separately for each roughages using in vivo and in situ procedure as a standard.
